About Lorlatinib
Lorlatinib or Lorbrena is an enzyme inhibitor approved for the treatment of individuals with metastatic anaplastic lymphoma kinase (ALK)-positive non-small cell lung cancer (NSCLC).
Lorbrena has progressed while being treated with crizotinib and at least one additional ALK inhibitor for metastatic disease; or alectinib as the first ALK inhibitor therapy for metastatic disease; or ceritinib as the first ALK inhibitor therapy for metastatic disease.

25 mg or 100 mg tablets
Recommended Dosage:
The recommended dosage is Lorlatinib 100 mg tablets orally once daily.
Important: If patients experience any adverse reactions from the Lorlatinib treatment, they must consult their treating doctor.
Warnings & Precautions
- Concurrent Use of Strong CYP3A Inhibitors Increases the Risk of Serious Hepatotoxicity. Before starting Lorlatinib, discontinue strong CYP3A inducers for three plasma half-lives.
- Seizures, hallucinations, changes in cognitive function, mood (including suicidal thoughts), speech, mental state, and sleep are instances of CNS effects. Withhold and restart Lorlatinib or Lorbrena at the same or reduced dosage, or discontinue Lorlatinib completely, depending on severity. It is necessary to consult the treating doctor for reduced, withhold, or discontinuation advice.
- Continue or increase the dose of lipid-lowering medications during the treatment (Hyperlipidemia). Withdraw and re-initiate Lorlatinib tablets at the same or reduced dosage according to severity.
- Withdraw and resume Lorlatinib or Lorbrena at the same or lower dose, depending on severity (Atrioventricular Block).
- Stop following Lorbrena treatment immediately in patients with suspected ILD/pneumonitis (Interstitial Lung Disease/Pneumonitis). For any severity of treatment-related ILD/pneumonitis, discontinue the Lorlatinib treatment.
- There are increased possibilities of fetal damage (Embryo-Fetal Toxicity). Inform females of the reproductive potential of the risk to a fetus. Men and women of reproductive age should be advised to follow an effective non-hormonal procedure of contraception.

Storage and Handling
To protect them from light, store the tablets between 20ºC to 25ºC. Excursions are permitted between 15ºC to 30ºC.
