About Brentuximab Vedotin or Adcetris
Brentuximab Vedotin is primarily used to treat lymphomas such as Hodgkin’s lymphoma and systemic anaplastic large cell lymphoma (sALCL). The packaging of Brentuximab injection comprises a 50mg vial for intravenous infusion.
Hodgkin lymphoma following autologous stem cell transplant (ASCT) failure or after at least two prior multi-agent chemotherapy regimens in patients who are not ASCT candidates.
Systemic anaplastic large cell lymphoma following at least one prior multi-agent chemotherapy treatment failure.
Strength: For injection,50 mg of brentuximab vedotin in a single-use vial as a sterile, white to off-white lyophilized, preservative-free cake or powder.
Recommended Dosage
The recommended dose for Adcetris is 1.8 mg/kg given as an intravenous infusion every 3 weeks. However, the dosage may vary based on the patient’s weight and medical conditions. It is advised to continue the treatment until the condition progresses or there are no serious brentuximab vedotin side effects visible in the patient. Dosage must be reduced for patients with mild hepatic impairment.

Warning & Precautions
- Concurrent usage with bleomycin is not recommended due to pulmonary harm.
- Patients must be monitored for neuropathy and implement appropriate dosage modifications.
- If anaphylaxis occurs, the infusion must be discontinued.
- If neutropenia of Grade 3 or 4 develops, the dosage must be reduced.
- Infection with bacteria, fungi, or viruses is common.
- Patients with rapidly growing tumors or a significant tumor burden must be closely monitored.
- Liver enzymes and bilirubin
- Discontinuation is recommended in cases of Stevens-Johnson syndrome or toxic epidermal necrolysis.
- Fetal harm is common in some cases. Pregnant women must be advised of the potential hazards to the fetus.

Use in Specific Populations
Usage of brentuximab must be avoided in cases of moderate or severe hepatic impairment or severe renal impairment as MMAE exposure and adverse reactions are increased.
Storage & Handling
Adcetris is a white to off-white cake or powder concentrate for infusion solution delivered in a glass vial. Each Brentuximab Vedotin pack contains one vial.
To protect from light, keep the vial in the refrigerator at 2-8°C (36-46°F) in its original carton.
